300: Rise of an Empire er fortalt i samme storslåede visuelle stil som blockbusteren 300, og er et nyt kapitel i denne episke saga fra oldtidens Grækenland. Denne gang er actionscenerne rykket til en ny slagmark – det åbne hav. Den athenske general, Themistokles (Sullivan Stapleton), der både er en erfaren kriger og en gudsbenådet strateg, forsøger at samle Grækenland mod den enorme angribende persiske hær, som er anført af den persiske konge Xerxes (Rodrigo Santoro), og af den hævn- og blodtørstige hærførerinde, Artemisia (Eva Green). Themistokles forsøger at overtale Spartas dronning Gorgo (Lena Headey) til at kæmpe med sig, men spartanerne kæmper kun for Sparta, siger hun, og har ingen drøm om et forenet Grækenland.
